I just got a flat tire on my rental car, I believe it happened close to my hotel, the tire is still pumped but sounds like it is losing air what is the best course of action?

If during working hours call the company. If after working hours call the emergency line that they gave and they will come and fix it for you.

Call on emergency contact provided by the rental car company. They might give you the best solution.
