No, you can take the E train to Lexington Avenue–53rd Street station. If you have luggage, there are elevators at both stations.
Only about 10 minutes away by subway. Take the A, C, or E train to 42nd Street–Port Authority Bus Terminal.

The 2 train goes to a separate station on the east side of Penn Station. There is no free transfer to the [A/C/E] trains from the [1/2/3] trains at Penn Station.

Weekends are off peak. You can actually buy a "city ticket" which is only for the weekends at half price of off peak if going to or coming from Manhattan.
There are over 25 departures to this specific spot daily from NJ Transit TRAINS. It is the North East Corridor line, and either go to NJ Transit to look up exact times, or if your in the area stop by and grab a detailed schedule for free, they're located all over the NJ Transit section of Penn Station

All depends on where your going or coming from. For example, a trip to MetroPark on Northeast CORRIDOR Nj Transit train, it takes about an hour and ten minutes, but to get to Mets-Willets Point on Port Washington line of LIRR it's about 15-20 minutes
There is an actually Amtrak section of Penn Station. The platforms change with every trip, must be there to find out.
