Probably best to take a cab. If you're traveling light and want to save some money you could take a bus or cab to 74th st and roosevelt Ave queens subway station and take the E train to 34th st.

I found the subway was best, time square traffic is tough for cabs/uber
I would take a cab. The uber and Lyft system in NYC isn't affordable. Also though just a heads up this hotel has Bed Bugs as of three weeks ago.
