It does not have a main entrance persay. There are a series of access points off of both 75 and 41 on the Western side of the peninsula. There are also a series of access points off of Turner River Road as well as bird on road and wagon wheel road. There is no real restriction on pedestrian access only motorized vehicle access. The Oasis visitor center may be the best place to explore and walk in on the Florida trail. This is about 2/3 of the way across from Miami to the West Coast on the North side of 41

It's like Robert said above, there is no main entrance there are just series of access points if you're going to enter it by a licensed and certified ORV (your vehicle must be inspected and stickered by the National Park Service for you to take it into the backcountry on the ORV trails) or you can walk in on foot anywhere or by canoe or other type of self-propelled watercraft providing water levels are appropriate.
