It depends on how much time you have... hours or days or... The Skyline Drive is through the Shenandoah National Park and spectacular - make sure you give yourself plenty of time. At about Waynesboro VA is where it ends and where the Blue Ridge Mountain Highway starts. There are lots of hiking trails though you must be prepared and reasonably fit.. You can do longer hikes and camp along the way - the Appalachian Trail, which starts up in Pennsylvania, wends it's way along through the mountains. Alternatively, you can spend time driving through the Valley, off the freeway, and visit the little towns dotted through the area.

Hike, drive the Skyline Drive and enjoy the overlooks, camp, see historic stuff in the area.
