Depends on the donation you do. A normal whole blood donation is 20 minutes tops, and doesn't really hurt past the initial needlestick (pretty much the same as getting a shot). Double red or platelet donations are longer and involve taking blood out, filtering it, and putting it back; they get more specialized donations that way (double red for trauma, platelets for cancer patients), but the donation is more difficult. Long answer short, yes, a little, but usually not very much, and it's a darn good thing to do.
