She should take Ferry Tour to see great city views from the river.
Just be aware that the real problem is getting back to the airport on time as well as getting through the subway down to a point of beginning... Subway trips from downtown take a long time. Be aware.

Yes she can but must rush back and forth from airport ASAP. A typical North or Southbound tour of Manhattan (NYC) takes about an hour to an hour and a half not including the cue to board double decker buses and train rides from and to JFK Airport (A, E trains and airport rail). if your wife can quickly get away to and from JFK Airport for about 6 - 8 hours she may successfully ride on both the North and Southbound guided Sightseeing Bus Tours. She shall start her Northbound tour(s) from the vicinity of Sightseeing's main office at 8th Avenue after buying an all day hop on/off pass online and picking up at that office. There she can get further information about where to take the Southbound leg which usually is a few blocks away from other tours. She must coordinate all transportation, walking, waiting, etc. into trip and add at least 1-2 hours for unforeseen delays, traffic, etc. May she enjoy sightseeing NYC.
