The correct answer to this question is yes. At a certain time the checkpoint closes and access to the inside terminal are emptied. In the morning, they reopen, but everyone must go through screening. This answer pertains to the old MSY airport and I do not know if this answer will apply after May 2019; in which the new MSY may open and different rules could or may apply.
