I paid off my car & they sent me the lien release letter. I need to get a paper title. As I understand, I only need to mail DMV the lien release letter and form TR-720B Manual Title Application. Is this info correct? What address do I mail these to?

Yes, and mail those to JoCo Treasury office.

The top of the form says drop it, the lien release and pay the fee at one of the Joco locations.
