How do they handle handicap, wheel chair seating, entrance and where is the seating area located

From what I saw, the handicap seating is at the top of the bottom level with a pretty wide area available. Everything was accessible by ramps.

Go to main entrance for Petco Park. There's a special entrance for wheelchair fans. Ask gate attendants where. Go to ground level elevator next to escalator. Go to top level...300 seating numbers level. From elevator walkways will lead you to seating. Ushers are there to assist handicap fans. They'll take you to your "seats". They'll assist you through the game to its end. ENJOY!

There are ramps throughout, and on front row of levels are situated wheelchairs/folding chairs.
