Can't speak to the 2001 Highlander, but with my 2003 Lexus ES300, the engine turned over, but the engine would NOT start. Once the key was associated with the vehicle, it worked just fine. You can see how to register your key with your make/model vehicle by doing a internet search. Mine was along the lines of 1. Key in ignition, turn to ON 2. Open/Shut driver door 10x 3. Fully depress and release Accelerator 5X 4. Turn Key to OFF 5. Remove key 6. Insert key and remove 3X 7. Insert known Good key 8. Car will chirp once 9. Remove Good Key 10. Insert New Key to be Coded 11. Turn Key to On Position (NOT Start) 12. Car will Chirp 3x when key is coded 13. Remove key Replace and repeat steps 10-12 for each key 14. Depress Brake to end cycle Your process may be similar.
