Adrian is right. I believe the tour was about an hour to an hour and a half. We took the historical tour in character, which happened to be for all museum members from some convention. That could have elongated ours but it felt like we were moving at the same speed as the tours behind us. However there is a nice display in the modern museum worth looking at before or after your tour, and of course a gift shop.

Tour was about 1.5 hours with the cub scouts. Adult parents and 9 and 10 year olds enjoyed it much. Some of the guides have tours in character. It was interesting. He was an iconic figure in American culture.
Ot was about the same for the regular tour.
You can spend one entire day and gou should come the next day and every single day during a week. !!! You can learn the whole Mark Twain History !!!
I think it depends on what type of person you are. The tour does take a little over an hour, there is usually a wait before your tour starts. There is a 15 minute film. There is also a section of the museum outlining his businesses and another section that talks about his family. Many people do not even go into those areas. If you are a plaque reader like then you will want to allow extra time for that. Don't forget the gift shop. I think 3-4 hours. Since you are already there you might want to consider seeing the Harriet Beecher Stowe house as well. A really great tour that focuses on the sociological and political impacts to our country. Which is typically another hour and a half. Sometimes when you go to the Mark Twain house the tour will be full for several hours seeing the Stowe during that time is a good use of the available time.
Would highly recommend taking the tour with the actors it really comes to life, takes about an hour and a half, then we walked the grounds for awhile!
