we have been quoted rates for 12 day cruise Royal Carribian out of Barcelona leaving in July. for 2 ocean view cabins, 2 persons in each of two cabins. If we elect to put 3 persons in one cabin and only 1 person in the other cabin effect the price?

Leave your cabin arrangements as is. You can't change bookings once made without penalties, unless you have travel insurance. You have the possibility to change sleeping arrangements yourself once on board.

Some cabins have a pull down bed . You would still need to book 2 and 2 and split the sleeping as you want
