Was there 2 Saturdays ago and there was a 20 min wait in the car. Was a nice day and a weekend, with 440+ manatee in the spring. Can't imagine that your wait would be any longer than mine.
What you can do is drive to the entrance on Blue Springs road and walk into the park and pay at the gate.

When i was there Friday it was about 10 minutes at the gate
Right now it is at least 1 hour wait (1pm on a Thursday)
