With respect to the rooms the doors are closed but they are never locked, so yes you are free to leave and go to the bathroom.
Many of the people working are watching you ready to give Clues and things...just let them know you have to go and they will help you out. Matt had to let me out during my last game. They do try to warn you before hand as well
