You would have to know someone working on the show. Unless a principal is out with illness/injury understudies, typically, only go on for weekday and matinee shows. That being said, try and get tickets for evening/weekend shows.

There is always a fee with ticketmaster but I don't know what it is. We but a subscription for 6 to 8 plays. I believe this is the most economical way to buy tickets.

Box office is open before shows, including in the evening. if you can, go on a night earlier in the week to buy your tickets shortly after that evening show begins, then there will be no line, and the night of your show you will not have to stand in line for tickets.

When I was there in January it appears that there is handicapped accessible seating. There is an elevator if your seats are in the balcony or above. There are washrooms on the balcony level but the space was small. There are washrooms downstairs that are spacious and plentiful. The advice given previously is sound advice and you should call ahead.
